Adaptogens in Supplements: Hype, Science, and Market Potential

Adaptogens — plants and fungi traditionally used to support resilience under stress — have experienced a remarkable resurgence. Once confined to Ayurvedic and Traditional Chinese Medicine, ingredients like ashwagandha, rhodiola, and lion’s mane are now headline actives in functional supplements sold globally.

Defining Adaptogens

The term “adaptogen” was coined in 1947 by Soviet pharmacologist Nikolai Lazarev to describe substances that increase non-specific resistance to stress. For a botanical to qualify, it must be non-toxic at normal doses, produce a normalising effect on physiological functions regardless of the direction of the stressor, and exert a broad influence on body systems rather than targeting a single organ.

Ashwagandha (Withania somnifera) remains the most clinically studied adaptogen, with a robust body of evidence supporting its effects on cortisol regulation, perceived stress, sleep quality, and physical endurance. Rhodiola rosea has demonstrated efficacy in reducing mental fatigue and improving cognitive performance under acute stress. Eleuthero (Eleutherococcus senticosus) has historically been used to support physical stamina and immune function.

What the Evidence Actually Says

Clinical data on adaptogens varies considerably in quality. Ashwagandha benefits from multiple double-blind, placebo-controlled trials with standardised KSM-66 and Sensoril extracts showing statistically significant reductions in serum cortisol and stress scores. Rhodiola evidence is promising but constrained by small sample sizes and heterogeneous extract standardisation across studies.

Lion’s mane (Hericium erinaceus) sits in an interesting position: preclinical data on nerve growth factor (NGF) stimulation and neuroprotection is compelling, but large-scale human trials are still limited. Nonetheless, it has captured enormous consumer interest as a nootropic, and early small-scale studies on cognitive function and mood support are encouraging.

Formulation and Dosing Considerations

Extract standardisation is critical to both efficacy and regulatory compliance. A supplement claiming to contain ashwagandha should specify the extract type (root, leaf, or whole plant), the standardisation parameter (e.g. ≥5% withanolides for KSM-66), and the daily dose used in supporting clinical studies. Underdosed or non-standardised extracts are unlikely to replicate study outcomes and risk misleading consumers.

Combining adaptogens is popular in stack formulations, but formulators must assess potential interactions — particularly with ingredients that affect HPA axis activity, thyroid function, or cytochrome P450 enzymes. A thorough safety dossier is essential before launch.

Regulatory Landscape in the EU

Most adaptogenic botanicals available in EU supplements are regulated under Directive 2002/46/EC. Health claims are tightly controlled: Regulation (EC) No 1924/2006 requires that all claims be on the EU Register of authorised claims. “Supports stress resilience” or “promotes relaxation” claims require robust substantiation and may need novel food authorisation for newer ingredients. Brands must ensure their safety assessment, labelling, and PIF documentation are complete before any EU market entry.
